    摘要:

    目的 探讨电针百会和大椎穴对颞叶癫痫大鼠海马组织中CA3和DG区ephrinA5的调控作用。方法 将30只Sprague-Dawley(SD)大鼠随机分为对照组、癫痫组和电针+癫痫组,每组各10只。建立氯化锂―匹罗卡品颞叶癫痫大鼠模型。造模成功的大鼠电针百会和大椎穴治疗8周后,分别取3组大鼠海马组织,采用实时荧光定量PCR(qRT-PCR)检测各组大鼠海马CA3和DG区ephrinA5 mRNA水平表达变化;采用Western blotting和免疫组织化学(免疫组化)法检测各组大鼠海马CA3和DG区ephrinA5蛋白水平表达变化。结果 qRT-PCR结果显示:与对照组相比,癫痫组大鼠海马组织中ephrinA5 mRNA表达下调(P <0.05)。通过8周电针百会和大椎穴连续治疗后,ephrinA5 mRNA水平上调(P <0.05)。Western blotting结果显示:ephrinA5蛋白水平变化趋势与mRNA水平相一致。免疫组化结果显示:在CA3区,癫痫组ephrinA5蛋白水平下调;电针后ephrinA5蛋白水平上调。而在DG区与对照组相比,癫痫组和电针+癫痫组,ephrinA5蛋白水平变化不明显。结论 电针百会和大椎穴的抗癫痫作用机制很可能与ephrinA5在海马CA3区中的调控机制密切相关。 [国际神经病学神经外科学杂志, 2021, 48(1): 50-54]

    Abstract:

    Objective To investigate the regulatory effect of electroacupuncture at Baihui and Dazhui points on ephrinA5 in the CA3 and DG regions of the hippocampal tissue in rats with temporal lobe epilepsy (TLE).Methods A total of 30 Sprague-Dawley rats were randomly divided into control group, TLE group, and electroacupuncture+TLE group, with 10 rats in each group. A rat model of TLE was established through lithium chloride-pilocarpine treatment. After successful modeling, the rats were subjected to electroacupuncture at Baihui and Dazhui points for 8 weeks; then, hippocampal tissue was collected from the rats in the three groups, and quantitative real-time PCR (qRT-PCR) was used to detect the changes in mRNA expression of ephrinA5 in the CA3 and DG regions of the hippocampus in the rats in each group; western blotting and immunohistochemistry were used to detect the changes in protein expression of ephrinA5 in the CA3 and DG regions of the hippocampus in the rats in each group.Results The results of qRT-PCR showed that the mRNA expression of ephrinA5 in the hippocampus tissue of rats in the TLE group was significantly down-regulated compared with that in the control group (P <0.05); after 8 consecutive weeks of electroacupuncture at Baihui and Dazhui points, the mRNA expression of ephrinA5 was significantly up-regulated (P <0.05). The results of western blotting showed that the changing trend of ephrinA5 protein was consistent with that of its mRNA. The results of immunohistochemistry indicated that the protein expression of ephrinA5 was significantly down-regulated in the CA3 region of rats in the TLE group; the ephrinA5 protein was significantly up-regulated after electroacupuncture. However, the protein expression of ephrinA5 in the DG region did not change obviously in the TLE group and electroacupuncture+TLE group compared with that in the control group.Conclusions The antiepileptic mechanism of electroacupuncture at Baihui and Dazhui points may be closely associated with the regulatory mechanism of ephrinA5 in the CA3 region of the hippocampus.
