    摘要:

    作为人体器官当中最为微妙的器官之一,脑组织受血脑屏障(BBB)的保护实现某些有害物质的侵入,但同时也限制了药物的组织摄取而无法实现药物疗效。虽然近几年分子神经科学发展突飞迅猛,但是因BBB的穿透限制而使许多新药无法达到预期效果,这也大大制约了新药开发进展。目前以被证实可提高跨BBB药物递送方式可分为:①化学传递如药物脂化等②生物学传递如吸附介导、载体介导、受体介导等③短暂性开发血脑屏障④“特洛伊木马”分子运载⑤纳米技术的应用如脂质体、纳米颗粒、聚合物胶束等。

    Abstract:

    Being the most delicate organ of the body, the brain is protected against potentially toxic substances by the blood-brain barrier (BBB), which restricts the entry of most pharmaceuticals into the brain. The developmental process for new drugs for the treatment of CNS disorders has not kept pace with progress in molecular neurosciences because most of the new drugs discovered are unable to cross the BBB。The various strategies that have been explored to increase drug delivery into the brain include ① chemical delivery systems②biological delivery systems③disruption of the BBB④the use of molecular Trojan horses⑤Nanomaterials for drug delivery across the blood–brain barrier.
