    摘要:

    目的 通过组织形态学和动物行为学观察、肝肾功能测定系统考察量子点及制备的量子点-Aβ-Ab探针的生物毒性作用,以此评价量子点及探针的生物相容性。方法 将正常C57BL小鼠30只随机分为量子点(QDs)组、量子点-Aβ-Ab(QDs-Aβ-Ab)组和对照组,每组10只,分别行量子点、量子点-Aβ-Ab探针、PBS侧脑室注射后,行HE染色、尼氏体染色、动物行为学观察及肝肾功能测定。结果 HE染色和尼氏染色示QDs组、QDs-Aβ-Ab组海马CA1区神经元形态正常,尼氏体清晰可见,与对照组无差异。行为学观察7 d,3组小鼠进食进水正常,二便正常,无举尾、无皮肤毛发颜色改变,无步态异常、无兴奋抑制反应,无死亡。QDs组和QDs-Aβ-Ab组血清ALT、AST、BUN和CRE水平与对照组无明显差异(P>0.05)。结论 量子点及量子点-Aβ-Ab探针对小鼠整体毒性作用小,生物相容性好。

    Abstract:

    Objective To study the toxicity of Quantum dots (QDs) and Quantum dots-Aβ-Ab (QDs-Aβ-Ab) probe to C57BL mice through histomorphological and behavioral comparisons, hepatic and renal function tests in order to evaluate the biocompatibility of QDs and QDs-Aβ-Ab probe.Methods Thirty normal C57BL mice were randomly divided into QDs, QDs-Aβ-Ab, and control groups.The responses were observed by behavioral comparisons, HE staining and nissl staining after injections of 5 μL QDs, QDs-Aβ-Ab probe or normil saline.Hepatic and renal functions were detected using automatic chemistry analyzer.Results Cell morphology, ultrastructure and nissl bodies in the hippocampal CA1 of the probe and the QDs groups were normal as the control group.Behavioral observation of seven days showed normal life behaviors and skin changes, walking disorders, excitatory linhibitory response or death did not occur in three groups.There were no significant differences in the contents of serum ALT, AST, BUN and CRE among the three groups.Conclusions The QDs and QDs-Aβ-Ab probes have no toxic effects and show a good biocompatibility in C57BL mice.
